Abstract: In this paper, has been described the synthesis of some novel tetrazolodiazepine derivatives starting from piperidin-4-ones, with a view to test the novel compounds for various biological activities. The structures of the synthesized compounds have been established by IR, 1H and 13C NMR, mass and elemental analysis. Representative compounds have been characterized by COSY, HETCOR and DEPT 135 spectra. The representative compounds of the tetrazolodiazepine have been tested for anti-inflammatory activity by carrageenan induced paw edema method. From the results of the in vivo study, it is observed that the compounds taken for the study possess fairly good activity. Among the synthesized compounds, 6,8-bis-(3-chlorophenyl)-5-methyl-6,7,8,9-tetrahydro-5H-tetrazolo[1,5-d][1,4]diazepine 3c shows potent anti-inflammatory activity.
