Abstract: 5,6,7,3-Tetramethoxy-4'-hydroxy-8-C-prenylflavone5 isolated from the leaves of Malaysian Orthosiphon stamineus has been synthesized by synthetic route. 2,4,5,6-Tetrahydroxy­aceto­phenone 1 on treatment with dimethyl sulphate affords 2-hydroxy-4, 5, 6-trimethoxyacetophenone 2 and several other minor compounds. Compound 2 on prenylation yields 2-hydroxy-4,5,6-trimethoxy-3-C-prenylacetophenone 3. Alkaline condensation of 3 and 3-methoxy-4-hydroxybenzaldehyde affords 2',4-dihydroxy-3,4',5',6'-tetramethoxy-8-C-prenylchalcone 4. 2,3-Dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) treatment of 4 furnishes the title compound 5.
