Abstract: Baclofen, a gamma-amino butyric acid (GABA) derivative is mostly used to treat spasticity, alcoholism and hiccups. It contains a potential impurity namely baclofen impurity-A as per Indian Pharmacopoeia and other official compendiums. The objective of the present study is to synthesize and characterize baclofen impurity-A with the help of sophisticated analytical techniques like Fourier-Transform Infrared Spectroscopy (FT-IR), Differential Scanning Calorimetry (DSC), Thermo-Gravimetric Analysis (TGA), Nuclear Magnetic Resonance (NMR), Liquid Chromatography Mass Spectrometry (LC-MS), Elemental Analyzer (CHN), High Performance Liquid Chromatography (HPLC) and Gas Chromatography Head Space (GC-HS). Results show that baclofen impurity can be obtained in the purest form (>99.0%) to provide a reference standard.
