Abstract: An efficient synthesis of intermediates for macrolide antifungals patulolide A 14 and pyrenophorin 16 are described with the key steps involving (i) coupling of the alkyl lithium with α,β-unsaturated aldehyde 8. (ii) the oxidation of the cis-trans mixture of γ-oxo-α,β- unsaturated aldehyde to the trans-γ-oxo-α,β-unsaturated carboxylic acid 12.
