Title: Synthesis and blood pressure lowering activity of some newer quinazolinones

Abstract: Some new 3-[3'-(aminoacetylthiosemicarbazido)-4'-oxo-2'-(substituted-aryl)-1'-thiazolidinyl] -2-methylmonosubstituted-quinazolin-4(3H)-ones 5a-j have been synthesized by 1,5 cyclocondensation of thioglycolic acid with 3-(aminoacetylthiosemicarbazidosubsti tutedarylidene)-2-methylmono-substitutedquinazolin-4(3H)ones 4a-j. All the newly synthesized compounds of the series have been evaluated for their hypotensive activity. The most active compound of the series is 3-[1'-(aminoacetylthiosemicarbazido)-4'-oxo-5'-(4-hydroxy ,3-methoxyphenyl)-1'-thiazolidinyl]-2-methyl-6-bromoquinazolin-4(3H)-one 5h, which has shown blood pressure lowering activity of 83 mmHg at a dose of 2.5 mg/kg i.v. The structures of all these compounds have been delineated by elemental analysis and spectral studies (IR and 1H NMR).
