Title: Pyramiding of chitinase and glucanase genes for fungal resistance

Abstract: Binary vector constructs with antifungal genes namely basic chitinase, basic glucanase and acidic glucanase with CaMV 35S promoter were synthesized. Since chitinase and glucanase have synergistic effect for fungal growth inhibition, chimeric constructs having both the genes were also made. Basic chitinase cDNA and basic glucanase cDNA with their individual 35S promoters were put together in BinAR 19. Besides this, acidic glucanase can release glucan oligomers from cell wall which act as elicitors for triggering defense response. To use this property of acidic glucanase in combination with chitinase mediated fungal cell wall hydrolysis, construct having basicĀ· chitinase and acidic glucanase was also made. Beside the better expected antifungal efficiency of these chimeric constructs in comparison to individual gene constructs, chances of resistant pathogen to evolve against two genes at a time will be quite reduced and protection will be longer lasting.
