Authors: Thiripurasundari, U; Rao, M V

Abstract: The present
study reports an efficient protocol for indirect shoot organogenesis and
plantlets regeneration of Coccinia grandis (L.) Voigt from nodal
explants. The maximum frequency of organogenic callus induction was observed in
MS medium supplemented with 0.1 mg/L 1-naphthaleneacetic acid (NAA)
and 1.0 mg/L N6-benzyladenine (BA). Multiple shoot induction was
achieved from the surface of callus on regeneration medium [MS nutrients
supplemented with NAA, BA and various concentrations of kinetin (KIN) 0.1-2.0
mg/L]. The highest shoot multiplication (90%) with the increased numbers of
shoots (8.7±1.8) and the shoot mean length (8.8±0.4 cm) was achieved on MS
medium with 0.1 mg/L NAA, 1.0 mg/L BA and 0.5 mg/L KIN. The regenerated shoots
were rooted in vitro on MS medium containing 0.1 mg/L IBA. Plantlets
with well developed shoot and root systems were successfully acclimatized
(83.5%) and exhibited normal morphology and growth characteristics.

Title: Somatic embryogenesis in Jatropha curcus L. using cotyledonary leaves

Authors: Saxena, Sonal; Sharma, Amit; Sardana, Jyoti; Sharma, Madan Mohan; Batra, Amla

Abstract: An efficient
protocol has been developed for high frequency plant regeneration through
somatic embryogenesis in the biodiesel plant, Jatropha curcus. Cotyledonary leaves were used for the development
of the embryogenic callus on MS medium with
0.2 mg/L indole-3-acetic acid (IAA), which developed within
3-4 wk. Later, somatic embryos developed on MS medium with IAA (0.2 mg/L) along
with 6-benzyl aminopurine (BAP; 1.5 mg/L). Fist, globular embryo clumps were
seen within 3-4 wk and then they got transformed into heart, torpedo and
cotyledonary shaped embryos on the same medium after 6 wk. On further
subculture on the same medium, embryos germinated with shoot and root
primordia, which ultimately gave rise to complete plantlets within 4-5 wk. The
pantlets were hardened and transferred to the soil. Around 50% plantlets
survived in the soil.
Authors: Ramadevi, T; Ugraiah, A; Pullaiah, T

Abstract: In vitro propagation
of Boucerosia diffusa Wight
(Apocynaceae), an endemic medicinal plant of Western Ghats, (Tamil Nadu, India) was
carried out. Nodal explants were cultured on MS medium supplemented with
cytokinins [benzyl aminopurine (BAP), kinetin (Kn), thidiazuron (TDZ) &
zeatin] either individually or in combinations. Shoot buds proliferated on MS
medium+BAP (8.87 µM) and repeated
subculture on MS+BAP (8.87 µM)+TDZ
(2.27 µM) promoted the shoot
multiplication. The regenerated shoots were then rooted on MS+NAA (2.69 µM). The micropropagated planlets were
hardened with the survival success rate of 56%. The developed micropropagation
protocol would help ex situ
conservation of this endemic species.
